Reconstitution is simple arithmetic once you have the three inputs. The concentration is the peptide mass divided by the water volume: a 10 mg vial in 2 mL of Bacteriostatic Water gives 5 mg/mL, or 5,000 mcg per millilitre. On a U-100 insulin syringe, where one unit is 0.01 mL, that works out to 50 mcg per unit.
The volume to draw for a chosen target amount is that target divided by the concentration. Adding more water spreads the same peptide across more liquid, so each unit carries less and small amounts become easier to measure. The aliquots per vial figure is the total peptide divided by your target, so you can see how far a vial goes before you order again.

That is a research choice, and the calculator exists to show the trade-off rather than prescribe an amount. Adding more water lowers the concentration, which makes small target amounts easier to measure accurately on a syringe; adding less concentrates the solution. On a standard U-100 insulin syringe, 100 units equal 1 mL, so one unit is 0.01 mL. The calculator converts your target amount into both units and millilitres so you can read whichever your syringe is marked in.
